Natural selection interacts with the local recombination rate to shape the evolution of hybrid genomes

While hybridization between species is increasingly appreciated to be a common occurrence, little is known about the forces that govern the subsequent evolution of hybrid genomes. We considered this question in three independent, naturally-occurring hybrid populations formed between swordtail fish species Xiphophorus birchmanni and X. malinche. To this end, we built a fine-scale genetic map and inferred patterns of local ancestry along the genomes of 690 individuals sampled from the three populations. In all three cases, we found hybrid ancestry to be more common in regions of high recombination and where there is linkage to fewer putative targets of selection. These same patterns are also apparent in a reanalysis of human-Neanderthal admixture. Our results lend support to models in which ancestry from the \"minor\" parental species persists only where it is rapidly uncoupled from alleles that are deleterious in hybrids, and show the retention of hybrid ancestry to be at least in part predictable from genomic features. Our analyses further indicate that in swordtail fish, the dominant source of selection on hybrids stems from deleterious combinations of epistatically-interacting alleles.\n\nOne sentence summaryThe persistence of hybrid ancestry is predictable from local recombination rates, in three replicate hybrid populations as well as in humans.

GADD45α regulates cell proliferation and DNA repair of BRL-3A cells that treated by FZD/UVC via P38, JNK, CDC2/CCNB1, AKT and MTOR pathways

GADD45 is a stress-induced gene activated by a variety of stress stimuli, including ultraviolet and ionizing radiation, and involved in cell cycle regulation, apoptosis, maintenance, genomic stability, DNA repair and immune response. However, the effects and regulatory mechanism of GADD45 on proliferation, apoptosis and DNA damage repair of hepatocytes in liver regeneration remains unclear. In this study overexpression of GADD45 significantly inhibited the cell viability, proliferation, the number of cells in G1 and S phases, and of furazolidone (FZD) or UVC induced apoptosis of BRL-3A cells and decreased the inhibition of FZD/UVC on the viability, proliferation of BRL-3A cells, while increased the number of cells in G2/M phase of BRL-3A cells and FZD/UVC induced S phase arrest. Downregulated GADD45 induced the viability, proliferation, the number of cells in S and G2/M phases and the inhibition of FZD/UVC on the viability, proliferation of BRL-3A cells increased, while decreased apoptosis, the number of cells in G1 phases of BRL-3A cells and FZD/UVC induced S phase arrest. The results of qRT-PCR and western blot showed that genes/proteins related to P38MAPK, JNK, CDC2/CCNB1, AKT and MTOR signaling pathways were significantly changed in normal BRL-3A cells. The expression profiles of cell cycle, proliferation and apoptosis related genes/proteins in FZD/UVC treated BRL-3A cells were also detected by qRT-PCR and western blot, and the results indicated that the expression of Myc, Bcl-2, Ccnd1, PCNA, P21, Ccnb1, Caspase3, Caspase8, Caspase9 and Bax have significantly changes.

Parsing Of Compositions And Microstructure Characteristics For Rust-Spots Of Pear Pericarp

The commercial value of Kurles pears pericarp, a popular and favored fruit for its unique aroma and refreshingly crisp texture, had sharply decreased because of a rust breakout of the beloved pear in China during the atmosphere-controlled storage. High performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LC-MS) were used to analyze rust spots on the pericarp of Kurle pears. Therefore, the chemical compounds of four various eluates, originating from the rust-colored substance collected from the pears pericarp effected, were identified successfully for the first time, which were just rhein, aloe-emodin, chrysophanol and emodin, respectively. Taken together with microstructure characteristics for rust-spots of Kurle pear pericarp, it was no doubt that these eluates were the main factors affecting the rust spots on the pericarp of the Kurle pears during the atmosphere-controlled storage, which was a sign and consequence resisting the undesirable stress of the external environment.

Carrion Fly-Derived DNA Metabarcoding Is An Effective Tool For Mammal Surveys: Evidence From A Known Tropical Mammal Community

Metabarcoding of vertebrate DNA derived from carrion flies has been proposed as a promising tool for biodiversity monitoring. To evaluate its efficacy, we conducted metabarcoding surveys of carrion flies on Barro Colorado Island (BCI), Panama, which has a well-known mammal community, and compared our results against diurnal transect counts and camera-trapping. We collected 1084 flies in 29 sampling days, which were pooled into 102 DNA extractions. We then conducted metabarcoding with mammal-specific (16S) and vertebrate-specific (12S) primers targeting mtDNA, and sequenced these amplicons on Illumina MiSeq. For taxonomic assignment, we compared BLAST with the new program PROTAX, and we found PROTAX significantly improved species identifications. We detected 20 mammal, four bird, and one lizard species from carrion fly metabarcoding, all but one of which were previously known from BCI. Fly metabarcoding detected more mammal species than concurrent transect counts (29 sampling days, 13 species) and concurrent camera-trapping (84 sampling days, 17 species), and detected 67% of the number of mammal species documented by 8 years of transect counts and camera-trapping combined, although fly metabarcoding missed several abundant species. This study demonstrates that carrion fly metabarcoding is a powerful tool for mammal biodiversity surveys, and has the potential to detect a broader range of species than more commonly used methods.
