Establishing RPTE-derived cell lines expressing hTERT for studying BK polyomavirus

Abstract

We previously established an infection model for BKPyV in primary human renal proximal tubule epithelial (RPTE) cells. Use of these cells is limited by their inability to be passaged extensively. We describe RPTE cells immortalized with hTERT, which can serve as a model system for acute or persistent BKPyV infection.

Tandem repeats in methanogenic archaea and their viruses reveal a shared potential for rapid genomic diversification

Methanogenic archaea drive global methane emissions, yet the molecular basis of their physiological flexibility is unclear. Archaeal adaptation is typically attributed to horizontal gene transfer, which is episodic and cannot explain rapid, reversible change. Here we show that direct tandem repeats (TRs) offer an intrinsic alternative. By screening 50,567 prokaryotic genomes, we find TR densities in Methanosarcina more than 30-fold above the prokaryotic average, paralleling Borg extrachromosomal elements. These TRs recur at conserved loci while sharing almost no sequence identity between strains, indicating independent origin, and evolve during routine cultivation. Methanogen TRs are transcribed, show ribosome occupancy, and lie near regulatory and cofactor-related genes, whilst being depleted around core energy-conserving and translational machinery. Genic TRs generate intrinsically disordered regions in proteins carrying nucleotide-binding, protein-protein interaction and cell-surface domains, and occur in structural and surface proteins of methanogen-infecting viruses. TRs are therefore an overlooked source of variability and adaptation potential in methanogens and their viruses.

DarHTG is a tripartite phage defence system for purine DNA base ADP-ribosylation

Bacteria evolved a variety of phage defence strategies to combat the constant onslaught of viral threats. DarTG toxin-antitoxin systems provide phage defence by ADP-ribosylation of bases in single-stranded DNA which interferes with viral replication and aborts phage infection. Here, we describe DarHTG, a system widespread among bacteria, that combines DarTG with a non-processive superfamily I helicase functioning as co-toxin for DarT and thereby enabling the ADP-ribosylation of double-stranded DNA. We identified two such DarHTG subclasses which catalyse the ADP-ribosylation of adenosine and guanosine bases, respectively, and reverse these modifications with structurally unique ARH superfamily antitoxins. Thus, DarHTG antitoxins expand the known macromolecular target spectrum of the ARH enzyme class to nucleic acids. These DarHTG systems confer resistance to a broad range of bacteriophages and can be triggered by the highly charged tails of viral single-stranded DNA-binding proteins, which occlude the active site of the ARH antitoxin.

Draft genome of Paraperlucidibaca sp. CMO1 isolated from marine oil snow in the Churchill Marine Observatory, MB, Canada

An oil-degrading enrichment culture was inoculated from Marine Oil Snow (MOS) collected during an oil-spill experiment at the Churchill Marine Observatory in winter 2024. The complete metagenome-assembled genome for Paraperlucidibaca sp. CMO1 was reconstructed, exhibiting the genetic potential to degrade hydrocarbons, including alkanes and aromatic compounds, as well as the ability to produce surfactants.
