Fe2+/H2O2-mediated oxidation of homogentisic acid indicates the production of ochronotic and non-ochronotic pigments. Implications in Alkaptonuria and beyond.

Homogentisic acid (HGA) can be oxidized by a combination of Fe2+ and H2O2 into a darkly colored high molecular mass pigment. Increasing the concentration of H2O2 can lead to the formation of a high molecular mass material that exhibits less absorbance in the visible range of the electromagnetic spectrum, while maintaining a strong absorbance in the UV range of the electromagnetic spectrum. FT-IR spectroscopy did indicate the presence of a chemical feature in the pigments generated through H2O2-mediated oxidation that is absent in pigments generated through air-mediated oxidation. Our observations could have implications in the pathophysiology of alkaptonuria. In alkaptonuria, patients suffer from homogentisic aciduria from birth, but develop ochronosis (darkening) of collagenous tissues much later in life due to the formation of a melanin-like pigment. Three major questions regarding ochronosis remain to be answered: 1) how is the pigment formed, 2) why does it appear by the third decade of life and 3) why is it sporadic in appearance? Our results suggest that ochronotic or non-ochronotic pigments can be generated from HGA depending on the oxidation reaction conditions. Thus, the absence of any visible pigment, as for younger alkaptonuria patients, could not necessarily mean the absence of HGA-derived melanin-like molecules. We compare our results and their potential implications for ochronosis to the changes in pigmentation observed in vitiligo or aging (greying) hair.

Gum Arabic promotes oxidation and ester hydrolysis.

We studied the interactions between gum Arabic and select catecholic compounds like caffeic acid, chlorogenic acid and catechol. We observed that GA is capable of promoting the auto-oxidation of the above-mentioned compounds into darkly colored pigments without the addition of redox-sensitive cations. Gum Arabic appeared to be unique among polysaccharide-based materials as many other types of polysaccharides promote the oxidation of the above-mentioned compounds only in the presence of redox cations like Fe2+ or Cu2+. RP-HPLC and SEC chromatographic techniques were employed to monitor the reactions and to observe the formation of high molecular mass, pigmented materials from gum Arabic and all three catecholic compounds. FT-IR spectroscopic analysis revealed that, despite their darkly colored appearances, the gum Arabic/pigment materials synthesized contain mostly gum Arabic and very little pigment. As chlorogenic acid is an ester of caffeic acid, we studied the capacity of gum Arabic to promote ester hydrolysis using acetylsalicylic acid as the model compound. We observed that gum Arabic did promote the hydrolysis of acetylsalicylic acid into salicylic acid. However, in all our experiments involving the pigment formation between gum Arabic and chlorogenic acid, we did not observe any evidence that chlorogenic acid was hydrolyzed leading to the release of caffeic acid during these reactions. In addition, we observed that heat treatment of gum Arabic did not affect its pro-oxidizing capability, but it did negatively affect its capability to hydrolyze salicylic acid. Thus, these two types of chemical reactivity present in the gum Arabic material may be associated with different components of the gum Arabic material.

Polysaccharide-mediated synthesis of melanins from serotonin and other 5-hydroxy indoles

As a continuation of our research on the melanin formation from catecholamines, we studied the polysaccharide-mediated oxidation of serotonin and other 5-hydroxy indoles into melanin-like materials. As for the catecholamines, we observed that many polysaccharides promote the oxidation of such compounds, particularly in the presence of Cu2+. The reactions were monitored using RP-HPLC and SEC techniques. Melanin-like materials were purified through dialysis and evaluated using UV_Vis and FT_IR spectroscopic techniques. One such material, synthesized from chondroitin sulfate type A and serotonin in the presence of Cu2+ was found to affect the release of IL-l{beta} and IL-6 cytokines from immune cells.
