Temporal regulation of TRP channels during partial sciatic nerve ligation is modulated by PLCγ in male mice

Neuropathic pain is a debilitating condition afflicting millions worldwide, still lacking a proper and effective treatment. Understanding the underlying mechanisms that lead to neuropathic pain may lead to the discovery of new targets. Previously, we found that PLC{square} is a key player in mechanical hypersensitivity triggered by either capsaicin or complete Freunds adjuvant induced inflammation. Here, we investigated the role of PLC{gamma} in neuropathic pain using a partial sciatic nerve ligation (PSNL) injury model in male mice and a peptide inhibitor of PLC{gamma} activation (TAT-pQYP) injected at different time points after injury. Mechanical hypersensitivity was reversed by TAT-pQYP at 7, 14, and 28 days post-injury (dpi). Furthermore, both TAT-pQYP and the TRPA1 inhibitor HC-030031, but not the TRPV1 inhibitor capsazepine, were able to reverse mechanical hypersensitivity at 14 dpi. In contrast, all three treatments significantly improved the mechanical threshold at 28 dpi. After a challenge with TRPV1 agonist capsaicin at 14 dpi, TAT-pQYP treated animals exhibited increased sensitivity. We also found decreased TRPV1 mRNA levels in the DRG of PSNL animals at 14 dpi that returned to baseline at 28 dpi. TAT-pQYP treatment normalized TRPV1 expression to sham levels at 14 days. Conversely, TRPA1 mRNA expression increased at 14 days and returned to baseline at 28 days, while TAT-pQYP reduced TRPA1 expression at 14 days. At this same time point, injection of a pan-PLC (U73122), or Trk (GNF-5837) inhibitor reduced mechanical hypersensitivity and decreased Trpa1 expression, whereas only PLC{square} inhibition by TAT-pQYP or Trk inhibition restored Trpv1 levels to Sham values. These findings indicate that PLC{gamma}, and Trk signaling are important for expression of Trpa1 and activity of Trpv1. Taken together, our results show that dynamic changes in Trpv1 expression in the PSNL model account for differential sensitivity to TRPV1 inhibition and suggest that clinically relevant pharmacological inhibitors targeting ion channels may vary in efficacy depending on the temporal regulation of expression during neuropathic pain development. Finally, we show that TAT-pQYP is able to reverse mechanical hypersensitivity arising from neuropathic pain, adding evidence that PLC{gamma} is a promising target to be explored for neuropathic pain management. Significance statementWe show that the efficacy of inhibiting TRPV1 or TRPA1 on neuropathic pain in a partial sciatic nerve ligation model and that pain mitigation depends on the dynamic changes of channel expression post-injury, in the dorsal root ganglion, emphasizing that temporal regulation of drug targets should be taken into account when choosing therapeutic strategies for pain treatment. Notably, we identify PLC{gamma} as a promising therapeutic target, as its inhibition with TAT-pQYP reverses mechanical hypersensitivity during different stages of injury progression by modulating expression and activity of TRP channels.

Antifungal susceptibility profile of clinically relevant species of the genus Sporothrix: establishment of Epidemiological Cutoff Values (ECVs) according to CLSI broth microdilution methodology

Sporotrichosis is a globally distributed subcutaneous mycosis caused mainly by Sporothrix brasiliensis, S. schenckii, and S. globosa. Cat-transmitted sporotrichosis, primarily caused by S. brasiliensis in South America and to a lesser extent by S. schenckii in Southeast Asia, is emerging as a significant public health concern, due its outbreak potential. Itraconazole is the first-choice drug for treatment of human and cats, but reduced susceptibility has been reported based on previously proposed epidemiological cut-off values (ECVs). To support resistance surveillance, we aimed to establish CLSI-endorsed ECVs for these clinically relevant Sporothrix species. A total of 3,588 minimum inhibitory concentration (MIC) values for seven antifungal agents (amphotericin B, itraconazole, posaconazole, voriconazole, isavuconazole, olorofim, and terbinafine) were obtained from 19 international laboratories. Four of seven antifungals met the CLSI M57 guidelines criteria to determine the ECV. Established ECVs for amphotericin B were found to be high with 8 {micro}g/mL for S. brasiliensis and S. globosa, and 4 {micro}g/mL for S. schenckii. Itraconazole ECVs were 4 {micro}g/mL for S. brasiliensis and S. schenckii. Posaconazole ECVs were 4 {micro}g/mL for all three species (tentative for S. globosa), while the terbinafine ECV for S. brasiliensis was 0.12 {micro}g/mL. Olorofim demonstrated good in vitro activity, particularly against S. brasiliensis. Overall, this study establishes validated ECVs for key antifungals against Sporothrix species and identifies a low prevalence of non-wild type (NWT) isolates, supporting ongoing antifungal resistance monitoring.

Pten is a disconnecting node in the molecular landscape of the proliferation/quiescence decision during mammary gland acinogenesis

Cell context is key for cell phenotype. Using physiologically relevant models of laminin-rich ECM (lrECM) induction of mammary epithelial cell quiescence and differentiation, we have provided a landscape of the status of key molecular players involved in the proliferation/quiescence decision. Repression of some positive regulators of the cell cycle, such as cyclins and CDKs, occurred already at the mRNA level, whereas negative regulators of the cell cycle, such as Pten and p27, were upregulated only at the protein level. Interestingly, cell cycle arrest occurred despite the active status of Fak, Src and PI3k, because their downstream proliferative signalling pathways were repressed, suggesting the existence of a disconnecting node between upstream and downstream proliferative signalling in quiescent cells. Pten fulfils this role. Inhibition of Pten increased proliferation and restored Akt/mTORC1/2 and Mapk signalling in cells exposed to lrECM. In mice, Pten levels were positively correlated to the basement membrane thickness in the developing mammary epithelia, and Pten localized to the apicolateral membrane of luminal cells both in in ducts and near the nascent lumen in terminal end bud, characteristics consistent with a role for Pten in inducing and sustaining quiescence and tissue architecture. Accordingly, in 3D acininogenesis models, Pten was required for the onset and maintenance of quiescence, cell polarity and lumen assembly. The notion that lrECM-triggered differentiation involves a signalling circuitry with many layers of regulation provides an explanation for the resilience of quiescence within a growth-suppressive microenvironment, and that perturbations in master regulators, such as Pten, could disrupt the quiescent phenotype.

Integrated metabolomic, molecular networking and genome mining analyses uncover novel angucyclines from Streptomyces sp. RO-S4 isolated from Bejaia Bay, Algeria

Multi-omic approaches have recently made big strides towards the effective exploration of microorganisms and accelerating the discovery of new bioactive compounds. We combined metabolomic, molecular networking, and genomic-based approaches to investigate the metabolic potential of the Streptomyces sp. RO-S4 strain isolated from the polluted waters of Bejaia Bay in Algeria. Antagonistic assays against methicillin-resistant Staphylococcus aureus with RO-S4 organic extracts showed an inhibition zone of 20 mm by the agar diffusion method, and its minimum inhibitory concentration was 16 g/mL. A molecular network was created using GNPS and annotated through the comparison of MS/MS spectra against several databases. The predominant compounds in the RO-S4 extract belonged to the angucyclines family. Three compounds were annotated as known metabolites, while all the others were putatively new to Science. Notably, all compounds had fridamycin-like aglycones, and several of them had a lactonized D ring analogous to that of urdamycin L. The whole genome of Streptomyces RO-S4 was sequenced to identify the biosynthetic gene cluster (BGC) encoding for these angucyclines, which yielded a draft genome of 7,497,846 bp with 72.4% G+C content. Subsequently, a genome mining analysis revealed 19 putative biosynthetic gene clusters, including a grincamycin-like BGC with a high similarity to that of Streptomyces sp. CZN-748 previously reported to also produce mostly open fridamycin-like aglycones. As the ring-opening process leading to these compounds is still not defined, we performed comparative analysis with other angucycline BGCs and advanced some hypotheses to explain the ring-opening and lactone formation, possibly linked to the uncoupling between the activity of GcnE and GcnM homologues in the RO-S4 strain. The combination of metabolomic and genomic approaches greatly improved the interpretation of the metabolic potential of the RO-S4 strain.

Chlamydia trachomatis outbreak: when the virulence-associated genome backbone imports a prevalence-associated major antigen signature

Chlamydia trachomatis is the most prevalent sexually transmitted bacteria worldwide and the causative agent of blinding trachoma. Strains are classified based on ompA genotypes, which are strongly linked with differential tissue tropism and disease outcomes. A lymphogranuloma venereum (LGV) epidemics, characterized by ulcerative proctitis, has emerged in the last two decades (mainly L2b genotype), raising high concern especially due to its circulation among men who have sex with men (MSM). Here, we report an ongoing outbreak (mostly affecting HIV-positive MSM engaging in high-risk practices) caused by an L2b strain with a rather unique genome makeup that precluded the laboratory notification of this outbreak as LGV due to its non-LGV ompA signature. Homologous recombination mediated the transfer of a ~4.5Kbp fragment enrolling CT681/ompA and neighboring genes (CT677/rrf, CT678/pyrH, CT679/tsf, CT680/rpsB) from a serovar D/Da strain likely possessing the typical T1 clade genome backbone associated with most prevalent genotypes (E and F). The hybrid L2b/D-Da strain presents the adhesin and immunodominant antigen MOMP (coded by ompA) with an epitope repertoire typical of non-invasive genital strains, while keeping the genome-dispersed virulence fingerprint of a classical LGV (L2b) strain. As previously reported for inter-clade ompA exchange among non-LGV clades, this unprecedented C. trachomatis genomic mosaic involving a contemporary epidemiologically and clinically relevant LGV strain may have implications on its transmission, tissue tropism and pathogenic capabilities. The emergence of such variants with epidemic and pathogenic potential highlights the need of more oriented surveillance strategies focused on capturing the C. trachomatis evolution in action.
