Spaceflight-relevant microgravity triggers a sublethal Parkinson's-like dopaminergic decline in human neurons and organoids.

Spaceflight stressors may increase Parkinsons disease (PD) risk, but microgravitys specific contribution to human dopaminergic (DA) vulnerability remains undefined. Here, we exposed human iPSC-derived midbrain DA organoids and differentiated SH-SY5Y neurons to simulated microgravity for 72 hours. This exposure reduced neurite outgrowth, eroded DA identity, and activated familial-PD mitochondrial kinases without depleting extracellular dopamine. We observed severe mitochondrial dysfunction--including membrane potential loss and respiratory suppression--coupled with global translational repression. Furthermore, a sublethal, pre-degenerative state emerged, characterized by the selective release of mitochondrial cell-free DNA without apoptotic activation. Proteomic profiling revealed striking convergence with human PD transcriptomes and astronaut blood, highlighting shared suppression of mitochondrial metabolism, ribosomal translation, and altered RNA splicing. Together, these findings establish simulated microgravity as a sufficient, non-toxin trigger of early PD-like DA dysfunction, providing a robust human model for investigating prodromal neurodegeneration.

Inflammatory memory disables a DDR1 degradation checkpoint to enable pancreatic cancer growth

Inflammation and stroma remodeling regulate pancreatic ductal adenocarcinoma (PDAC), but how or if these cues are integrated at the molecular level remains unclear. Here, we identify a metabolic checkpoint that controls the stability of the collagen receptor DDR1, and subsequent tumorigenesis. We show that defective Col-I remodeling deprives PDAC cells of the high affinity DDR1 ligand, [3/4]Col-I, resulting in reduced ATP and activation of AMPK. AMPK phosphorylates DDR1 at T519, promoting its recognition by the E3 ubiquitin ligase adaptor FBXW2 and subsequent degradation. Importantly, this degradation pathway can be disabled by inflammatory signaling. Exposure to inflammatory cytokines induces methylation-dependent silencing of FBXW2, which establishes an inflammatory memory that preserves DDR1 stability, enabling sustained ligand-triggered receptor oligomerization and downstream NF-{kappa}B-NRF2 signaling even in restrictive stromal environments. Together, these findings identify regulated receptor turnover as a mechanism through which stromal architecture, metabolic state, and inflammatory memory are integrated to control PDAC progression.

Spatiotemporal dynamics and migration pathways of the transboundary Beet Webworm Loxostege sticticalis (Linnaeus) between Northern China and Mongolia in 2022

Clarifying the population dynamics, source-landing relationships, and migration pathways of the beet webworm Loxostege sticticalis (Linnaeus) both domestically and internationally, as well as understanding the meteorological mechanisms shaping these patterns, is pivotal for remote, accurate, and location-specific pest forecasting. Based on light trap data from northern China and field survey data from Mongolia in 2022, we simulated the migration trajectories, source regions, and primary landing areas of L. sticticalis, and analyzed the synoptic systems and processes during its migration. The results indicate frequent exchange of L. sticticalis populations between China and Mongolia in 2022. The L. sticticalis migrants initiating their flights from Mongolia primarily undertook a southeastward migration pathway, supplemented by eastward cyclonic and southwestward paths. The main landing areas were located in North China and Northeast China, with migration events potentially extending to Shandong, Heilongjiang, and Xinjiang provinces. Populations originating from North China exhibited a capacity for migrating into Northeast China and Mongolia through 1 to 5 consecutive nights of flight. During this period, the Northeast China Cold Vortex (NCCV) and the Mongolian Cyclone alternately regulated the synoptic circulation pattern governing the migration of L. sticticalis. The spatiotemporal distributions and intensities of these systems were key determinants of the transboundary migration routes and distances of L. sticticalis. The NCCV dominated, and the precipitation and downdrafts it induced were crucial for the massive landing of L. sticticalis in northern China.

Distinct contributions and sensorimotor encoding of PV and FoxP2 neurons in the external globus pallidus during perceptual decision-making

The external globus pallidus (GPe), a central nucleus of the basal ganglia (BG), comprises diverse cell types, including PV and FoxP2 neurons. While their roles in movement regulation and sensory processing have been studied, their contributions to perceptual decision-making remain unclear. Using a Go/No-Go visual task in mice, we found that optogenetic activation of either population impaired behavioral performance, whereas only inhibition of PV neurons, but not FoxP2 neurons, disrupted behavior. Electrophysiological recordings revealed that PV and FoxP2 neurons exhibited distinct encoding of visual stimuli, licking movement, and trial outcomes, with a larger fraction of PV neurons positively encoding the reward-predicting (Go) stimulus and the population overall preferring the rewarded over unrewarded outcome. FoxP2 neuron inhibition had minimal effects on spontaneous firing of neurons in the substantia nigra pars reticulata (SNr), a major BG output. By contrast, PV neuron inhibition altered both spontaneous and task-related activity of SNr neurons, reducing their preference for reward-predicting stimulus or unrewarded outcome in a subpopulation-specific manner. These findings indicate that PV and FoxP2 neurons in the GPe differentially influence sensorimotor processing and SNr activity, with PV neurons playing a key role in supporting accurate performance during perceptual decision-making.

Correlation Analysis of Breast Cancer Molecular Characteristics and Epithelial-Mesenchymal Transition in Circulating Tumor Cells: Based on Clinical Cases

BackgroundThis study aimed to investigate the correlation between epithelial-mesenchymal transition (EMT) in circulating tumor cells (CTCs) from peripheral blood and molecular characteristics of breast cancer. MethodsWe enrolled 48 breast cancer patients who underwent CTC testing at Shenzhen Peoples Hospitals Breast Surgery Department between September 2022 and May 2024. Intraoperative tissue samples underwent routine pathological and immunohistochemical examination. CTCs were detected using Canpatrol technology. Clinical data were analyzed statistically using SPSS 27.0. Categorical data were presented as percentages/frequencies. Comparisons used chi-square tests, with Yates correction for expected frequencies <5 but [≥]1. Statistical significance was set at P<0.05 (=0.05). ResultsEMT status showed no significant differences between ER-positive/negative, PR-positive/negative, HER2-positive/negative, Ki-67 [≥]14%/<14%, tumor size, or lymph node metastasis groups (P>0.05). Luminal-type tumors exhibited lower EMT grades than other subtypes, while triple-negative breast cancer (TNBC) showed higher EMT grades (P<0.05). ConclusionEMT grade correlates with luminal-type and TNBC subtypes, suggesting predictive value for these breast cancer classifications.

Glycyrrhizin inhibits LPS-induced inflammatory responses in goat ruminal epithelial cells in vitro

A long-term of high concentration feeding in ruminants can bring huge economic profits, but it also impose ruminants into great threat of suffering subacute ruminal acidosis (SARA). SARA is a kind of disease which attenuate the health, feed intake and production of ruminants, and when ruminants suffer SARA, the concentration of lipopolysaccharide (LPS) increase largely. Glycyrrhizin is reported to have anti-inflammation effects, and the study was conducted to investigate effects of glycyrrhizin on LPS-induced goat ruminal epithelial cells (GRECs) to provide evidence for using glycyrrhizin as a treatment for SARA. Effects of LPS, and glycyrrhizin on cell viability of GRECs were investigated, respectively. Then GRECs were stimulated with LPS (50 mg/L) for 2 h, and glycyrrhizin were added at the concentration of 0, 50, 75, 100 and 125 mg/L for 24 h to investigate the expression of inflammatory cytokines (by Elisa kits), the mRNA expression of NF-κB and inflammatory cytokines (by qRT-PCR), the distribution of Zo-1 and Occludin (by immunofluorescence staining), the expression of Occludin (by Western blot analysis), and the morphology of GRECs. The results showed that: (1) Glycyrrhizin at the concentration of 50, 75, 100, and 125 mg/L had no cytotoxic effects on GRECs, and LPS at the concentration of 50 mg/L significantly decreased the cell viability of GRECs. (2) Glycyrrhizin attenuated the expression and relative mRNA expression of TNF-α, IL-1β, IL-6, IL-8 and IL-12 by a dose-dependent manner, and significantly attenuated the relative mRNA expression of NF-κB. (3) Immunofluorescence staining and Western blot analysis showed that the quantity of Zo-1 and Occludin, and the expression of Occludin all increased with the treatment of glycyrrhizin. (4) Glycyrrhizin attenuated LPS-induced autophagy and protected the structural integrity of GRECs. In conclusion, glycyrrhizin significantly inhibited the inflammatory response in LPS-stimulated GRECs, and it may be used as a potential agent for the treatment of SARA.View Full Text

Comparative Genome Analysis of Scutellaria baicalensis and Scutellaria barbata Reveals the Evolution of Active Flavonoid Biosynthesis

Scutellaria baicalensis and Scutellaria barbata, common medicinal plants of the Lamiaceae family, produce specific flavonoid compounds with antioxidant and antitumor activities, including baicalein, scutellarein, norwogonin, wogonin, and their glycosides. Here, we reported two chromosome-level genome assemblies of S. baicalensis and S. barbata with significant quantitative chromosomal variation (2n = 18 and 2n = 26, respectively). The divergence of S. baicalensis and S. barbata occurred far earlier than previously reported, and a whole-genome duplication event was identified. The insertion of long terminal repeat elements after speciation might be responsible for the observed chromosomal expansion and rearrangement. The comparative genome analysis of congeneric species elucidated the species-specific evolution of chrysin and apigenin biosynthetic genes, such as the S. baicalensis-specific tandem duplication of the phenylalanine ammonia lyase (PAL) and chalcone synthase (CHS) genes, and the S. barbata-specific duplication of 4-CoA ligase (4CL) genes. In addition, the paralogous duplication, collinearity, and expression diversity of CYP82D subfamily members revealed the functional divergence of flavone hydroxylase genes between S. baicalensis and S. barbata. These Scutellaria genomes highlight the common and species-specific evolution of flavone biosynthetic genes, promoting the development of molecular breeding and the study of the biosynthesis and regulation of bioactive compounds.

Effects of different dietary NFC/NDF on rumen microbiomes of Karakul sheep based on Three Generations of Full-length Amplifiers sequencing

An study was was conducted to investigate the effects of dietary(non fibrous carbohydrate) NFC/(neutral detergent fiber)NDF on ruminal bacteria in Karakul sheep. Twelve Karakul sheep were assigned randomly to four dietary treatments of NFC/NDF (0.78, 1.23, 1.61 and 2.00 respectively) as group 1, 2, 3 to 4. The experiment lasted for four periods, period I (1~18 d), II (19~36 d), III (37~54 d) and IV (55~72 d). Ruminal digesta were collected consecutively for three days to measure pH and bacteria per period. The results indicated that the average ruminal pH and amounts of OTUs were decreased with the increase of dietary NFC/NDF for four periods. At phylum level, Bacteroidetes and Firmicutes were the predominant bacteria of four periods, Bacteroidetes were decreased, while the relative abundance of Firmicutes was increased with dietary NFC/NDF for four periods, but the difference wasnt significant (P>0.05). At genus level, the most relative abundance genus was unidentified-Lachnospiraceae which reached the highest in group 3 for four periods, but the difference wasnt significant (P>0.05). Conclusion: ruminal pH and bacteria were decreased with the increase of dietary NFC/NDF and the most dominant bacteria were not change with dietary NFC/NDF and periods in Karakul sheep.
