The impact of musical expertise on disentangled and contextual neural encoding of music revealed by generative music models

Music perception involves the intricate processing of individual notes and their contextual relationships within a piece. However, how the brain encodes and organizes these features, particularly in relation to musical expertise, remains unclear. Using noninvasive and invasive electrophysiological recordings alongside generative music models, we reveal that musicians exhibit neural encoding that is more attuned to the separation and integration of complex musical structures, with a pronounced left-hemispheric bias compared to non-musicians. Invasive recordings further highlight a hierarchical and spatially organized representation of musical features and context across brain regions. These findings advance our understanding of how the brain processes music and the role of musical training in shaping auditory cognition.

Binding of Calcium and Magnesium to Cardiac Troponin C

Cardiac troponin C (cTnC) is the Ca2+-sensing component of the thin filament. It contains structural sites (III/IV) which bind both Ca2+ and Mg2+, and a regulatory site (II) that has been thought to bind only Ca2+. The latter binding initiates a series of conformational changes that culminate in force production. We have quantified the interaction between site II and Ca2+/Mg2+ through Isothermal Titration Calorimetry and Thermodynamic Integration simulations. Direct and competitive binding titrations using wild type and a double mutant that significantly reduces binding to site II demonstrated that physiologically relevant concentrations of both Ca2+/Mg2+ interact with the same locus. Cytosolic free Mg2+ (~1 mM) could occupy a significant population of available site II, as this concentration of Mg2+ decreased the affinity for Ca2+ 1.4-fold. Interaction of Mg2+ with site II of cTnC likely has important functional consequences for the heart at baseline and in diseased states which decrease or increase availability of Mg2+ such as secondary hyperparathyroidism or ischemia, respectively.

High-resolution cryo-EM using beam-image shift at 200 keV

Recent advances in single-particle cryo-electron microscopy (cryo-EM) data collection utilizes beam-image shift to improve throughput. Despite implementation on 300 keV cryo-EM instruments, it remains unknown how well beam-image shift data collection affects data quality on 200 keV instruments and how much aberrations can be computationally corrected. To test this, we collected and analyzed a cryo-EM dataset of aldolase at 200 keV using beam-image shift. This analysis shows that beam tilt on the instrument initially limited the resolution of aldolase to 4.9[A]. After iterative rounds of aberration correction and particle polishing in RELION, we were able to obtain a 2.8[A] structure. This analysis demonstrates that software correction of microscope aberrations can provide a significant improvement in resolution at 200 keV.
