Musical Aesthetic Experience Reveals Differential Effects of Professional Vocal Training on Female College Students' Brain Function

Musical aesthetic experience can reveal students' cognitive, aesthetic, and emotional responses to music. However, how professional vocal training alters brain functions in female college students with varying levels of musicianship to promote these is unknown. Here we applied fMRI, psychological measures and Goldsmiths musical sophistication index (Gold-MSI) assessments to study 27 non-music major female college students consisting of instrument group (normal pitch accuracy with instrumental training), general group (normal pitch accuracy without instrumental training) and poor-pitch group (poor pitch accuracy and without instrumental training). After 1 year of one-on-one instructional practice, we found that professional vocal training enhanced information processing related to musical cognition and pleasure during musical aesthetic experiences across all three groups, involving improvements in motor control of vocalization, auditory feedback processing, musical memory and attention together with musical pleasure. Specifically, instrument group showed activation of reward circuits, general group exhibited activation of laryngeal motor control circuits, and poor-pitch group displayed activation of sensory/sensorimotor/motor control circuits. Both pre- and post-training, the instrument group exhibited higher connectivity than the other two groups in the left amygdala-dorsal anterior cingulate cortex (dACC) and the right posterior superior temporal gyrus (pSTG)-left insula pathway. After professional vocal training, the general group showed higher connectivity in the left amygdala-dACC, whereas the poor-pitch group demonstrated increased connectivity in the right pSTG-left insula and dACC-nucleus accumbens pathways. Surprisingly, professional vocal training activated the bed nucleus of the stria terminalis (BNST), a subcortical emotion center, and connectivity with the medial prefrontal cortex associated with musical information processing. Thus, vocal training can significantly improve musical cognition and aesthetic pleasure, and facilitates neural modulation in the brain's emotional centers.

Plant regulator of flower bud differentiation in in vitro plants of Cymbidium tortisepalum var. longibracteatum with TDZ as the key initiator

BackgroundCymbidium tortisepalum var. longibracteatum is a Class II nationally protected endangered plant in China with significant economic value. AimThis study aimed to establish an efficient in vitro flowering system and identify key hormonal factors regulating flower bud differentiation. MethodsOrthogonal experiments and hormone treatments were performed to evaluate the effects of TDZ, 6-BA, NAA, IBA, PP333, ABA, and GA3 on flower bud induction in tissue-cultured plantlets. ResultsTDZ was identified as the key initiator of flower bud differentiation, as no flower buds were induced in TDZ-free treatments. The optimal hormone combination was 1.0 mg{middle dot}L-1 6-BA + 0.4 mg{middle dot}L-1 TDZ + 0.8 mg{middle dot}L-1 NAA + 1.6 mg{middle dot}L-1 IBA, achieving a flower bud induction rate of 23.33% and a normal flower bud rate of 20.00%. PP333 inhibited flower bud differentiation but reduced malformation; at 0.2 mg{middle dot}L-1 PP333, the normal flower bud rate reached 20.00%. GA3 pretreatment resulted in a flower bud induction rate of 12.04%, whereas ABA pretreatment showed no significant promoting effect on flower bud induction. ConclusionsThis study provides an efficient in vitro flowering system and key hormonal parameters for shortening the breeding cycle and elucidating orchid flowering mechanisms.

Age-Associated Remodeling of neuroglial Connexin43 in the Mouse and Human Brain

Age-dependent changes in the major neuroglial junctional channel Connexin43 (Cx43) are poorly defined. We integrated public multi-organ and single-cell datasets with high-resolution morphological, biochemical, and functional analyses of human and mouse brains. We reveal a non-linear aging trajectory for Cx43, marked by an adaptational midlife elevation followed by a late-life decline. These molecular changes proceed in the absence of significant cell loss but are associated with extensive glial remodeling. Notably, while astrocytic gap junction coupling in the hippocampus remains largely preserved, aging induces a selective increase in hemichannel mediated dye uptake in both microglia and hippocampal astrocytes. In human cortical and hippocampal tissues, we confirm that aging drives a significant reduction in astrocytic Cx43 expression, alongside characteristic morphological simplification of cell structure and domain contraction. Together, our findings redefine the aging brain as a state of active, multi-level glial remodeling rather than a simple decline in connexin-mediated communication.

A humanized ossicle model of myelofibrosis reveals THPO-driven fibrosis, osteosclerosis and SPP1-dependent microenvironmental remodeling

Myelofibrosis (MF) is the most severe myeloproliferative neoplasm. Current therapies - except for allogeneic stem cell transplantation - are largely supportive which highlights the need for improved disease models and novel therapeutic targets. Here, we established a humanized MF model by transplanting thrombopoietin (THPO)-overexpressing human bone marrow CD34{square} cells into humanized bone marrow ossicles generated in immunodeficient NSG mice. THPO overexpression induced progressive reticulin fibrosis in vivo, accompanied by myeloid skewing, increased megakaryocyte clustering, and redistribution of human hematopoietic cells to murine spleen and femur, consistent with extramedullary hematopoiesis. THPO-driven ossicles also exhibited features of osteosclerosis, including increased trabecular bone and osteoid formation, indicating active pathological remodeling of the niche. Mechanistically, fibrosis was associated with increased SPP1/OPN expression, which was also observed in bone marrow biopsies from MF patients. Importantly, in vivo neutralization of SPP1 attenuated myeloid skewing, reduced megakaryocyte expansion, and decreased fibrosis severity, highlighting SPP1-driven niche remodeling as a potential therapeutic target in MF. This humanized MF model thus provides a translationally relevant platform to dissect microenvironment-driven MF pathogenesis and evaluate targeted therapies.

Prediction of range expansion and estimation of dispersal routes of water deer (Hydropotes inermis) in the transboundary region between China, the Russian Far East and the Korean Peninsula

Global changes may direct species expansion away from their current range. When such an expansion occurs, and a species colonizes a new region, it is important to monitor the habitat used by the species and use the information for updated management strategies. Water deer is listed as Vulnerable species in IUCN Red List and restricted to east central China and the Korean Peninsula. Since 2017 water deer has expanded its range towards northeast China and the Russian Far East. Our research focuses on data collected in northeast China and the Russian Far East during 2017-2021, with the purpose of providing support for a better understanding of habitat use and provide conservation suggestions. We used MaxEnt to model species niche and distribution and predict habitat suitability for water deer and applied the circuitscape to determine possible dispersal routes for the species. There is good quality habitat for water deer in the boundary area of the Yalu and Tumen River estuaries between China, North Korea, and the Russian Far East, as well as the east and west regions of the Korean Peninsula. Elevation, distance to cropland and water sources, and presence of wetlands were the variables that positively contributed to modelling the suitable habitats. Two possible dispersal routes were determined using the circuit theory, one was across the area from North Korea to the downstream Tumen transboundary region, and the other was across North Korea to the boundary region in China and along the tiger national park in northern China. A series of protected areas in North Korea, China, and Russia may support the dispersal of water deer. The establishment of a Northeast Asia landscape conservation network would help establish monitoring and conservation planning at a broad scale, and this study provides an example for the need for such a network

Isolation, screening, degradation characteristics of a quinclorac-degrading bacteria D and its potential of bioremediation for rice field environment polluted by quinclorac

Quinclorac (QNC) is a highly selective, hormonal, and low-toxic herbicide with a long duration. And the growth and development of subsequent crops are easily affected by QNC accumulated in the soil. In this paper, a QNC-degrading strain D was isolated and screened from the rice paddy soil. Through morphology, physiological and biochemical tests and 16Sr DNA gene analysis, strain D was identified as Cellulosimicrobium cellulans sp. And the QNC degradation characteristics of strain D were studied. Under the optimal culture conditions, the QNC-degrading rate was 45.9% after culturing for 21 days. The QNC-degrading efficiency of strain D in the field was evaluated by a simulated pot experiment. The results show that strain D can promote the growth of rice and QNC-degrading effectively. This research could provide a new bacterial species for microbial degradation of QNC and lay a theoretical foundation for further research on QNC remediation. ImportanceAt present, some QNC-degrading bacteria have been isolated from different environments, but there are no reports of Cellulosimicrobium cellulans sp. bacterial that could degrade QNC. In this study, a new QNC-degradation strain was selected from the paddy soil. The degradation characteristics of strain D were studied in detail. The results shown that strain D had a satisfactory quinclorac-degrading efficiency. Two degradation products of QNC by strain D were identified by HPLC-Q-TOF/MS: 3-pyridylacetic acid (138.0548 m/z) and 3-ethylpyridine (108.0805 m/z), which have not been reported before. The strain D had a potential ability of quinclorac-degrading effectively in the quinclorac-polluted paddy field environment.

Clinically-Driven Design of Synthetic Gene Regulatory Programs in Human Cells

Synthetic biology seeks to enable the rational design of regulatory molecules and circuits to reprogram cellular behavior. The application of this approach to human cells could lead to powerful gene and cell-based therapies that provide transformative ways to combat complex diseases. To date, however, synthetic genetic circuits are challenging to implement in clinically-relevant cell types and their components often present translational incompatibilities, greatly limiting the feasibility, efficacy and safety of this approach. Here, using a clinically-driven design process, we developed a toolkit of programmable synthetic transcription regulators that feature a compact human protein-based design, enable precise genome-orthogonal regulation, and can be modulated by FDA-approved small molecules. We demonstrate the toolkit by engineering therapeutic human immune cells with genetic programs that enable titratable production of immunotherapeutics, drug-regulated control of tumor killing in vivo and in 3D spheroid models, and the first multi-channel synthetic switch for independent control of immunotherapeutic genes. Our work establishes a powerful platform for engineering custom gene expression programs in mammalian cells with the potential to accelerate clinical translation of synthetic systems.

Primary mammary organoid model of lactation and involution

Mammary gland development occurs mainly after birth and is composed of three successive stages: puberty, pregnancy and lactation, and involution. These developmental stages are associated with major tissue remodeling, including extensive changes in mammary epithelium as well as surrounding stroma. Three-dimensional (3D) mammary organoid culture has become an important tool in mammary gland biology and enabled invaluable discoveries on pubertal mammary branching morphogenesis and breast cancer. However, a suitable 3D organoid model recapitulating key aspects of lactation and involution has been missing. Here, we describe a robust and straightforward mouse mammary organoid system modeling lactation and involution-like process, which can be applied to study mechanisms of physiological mammary gland lactation and involution as well as pregnancy-associated breast cancer.

Broadly Conserved Roles of TMEM-131 Family Proteins in Intracellular Collagen Assembly and Secretory Cargo Trafficking

Collagen is the most abundant protein in animals. Its dysregulation contributes to ageing and human disorders including tissue fibrosis in major organs. How premature collagens in the endoplasmic reticulum (ER) assemble and route for secretion remains molecularly undefined. From an RNAi screen, we identified an uncharacterized C. elegans gene tmem-131, deficiency of which impairs collagen production and activates ER stress response. TMEM-131 N-termini contain bacterial PapD chaperone-like (PapD-L) domains essential for collagen assembly and secretion. Human TMEM131 binds to COL1A2 and TRAPPC8 via N-terminal PapD-L and C-terminal domain, respectively, to drive collagen production. We provide evidence that previously undescribed roles of TMEM131 in collagen recruitment and secretion are evolutionarily conserved in C. elegans, Drosophila and humans.

OxyR senses reactive sulfane sulfur and activates genes for its removal in Escherichia coli

Reactive sulfane sulfur species such as hydrogen polysulfide and organic persulfide are newly recognized as normal cellular components, involved in signaling and protecting cells from oxidative stress. Their production is extensively studied, but their removal is less characterized. Herein, we showed that reactive sulfane sulfur is toxic at high levels, and it is mainly removed via reduction by thioredoxin and glutaredoxin with the release of H2S in Escherichia coli. OxyR is best known to respond to H2O2, and it also played an important role in responding to reactive sulfane sulfur under both aerobic and anaerobic conditions. It was modified by hydrogen polysulfide to OxyR C199-SSH, which activated the expression of thioredoxin 2 and glutaredoxin 1. This is a new type of OxyR modification. Bioinformatics analysis showed that OxyRs are widely present in bacteria, including strict anaerobic bacteria. Thus, the OxyR sensing of reactive sulfane sulfur may represent a conserved mechanism for bacteria to deal with sulfane sulfur stress.
