Enumeration, Antagonism and Enzymatic Activities of Microorganisms Isolated from Railway Station Soil

Soil is the well-known hotspot for microbial diversity. Therefore, for our investigation, we isolated, characterized, and identified microorganisms from railway station soil. Sampling was done subsequently after every 15 days interval, and from two different soil depths i.e. 0-15 cm and below during March to May of 2013. Further, soil isolates were examined for their antagonistic activity, against four soil born plant pathogens namely, Rhizoctonia solani, Aspergillus niger, Fusarium f sp pisi, and Sclerotinia sclerotiorum. Subsequently, isolates were screened for the presence of amylases, proteases, lipases and cellulases. For each interval of soil sampling, a gradual reduction in the microbial count was noticed from month March to May. Mucor species was observed only in the rainy days. The most promising enzymes producers were Bacillus sp., Aspergillus and Penicillium sp. Overall, the fungal isolates were better producers of enzymes as compared to bacterial isolates.

Characterization for Drought Tolerance and Physiological Efficiency in Novel Cytoplasmic Male Sterile Sources of Sunflower (Helianthus annuus L.)

AbstractSunflower is sensitive to drought and its hybrids have a limited cytoplasmic diversity. The wild cytoplasmic sources of sunflower are not well exploited to their potential for drought tolerance and hybrid development. In this respect, we carried out a Line x Tester based genetic study using 19 sunflower genotypes representing, 13 cytoplasmic male sterile (CMS) lines from wild and conventional sources, 2 maintainer lines, and 4 restorer lines. The CMS and maintainer lines were crossed with restorer lines to develop sixty one-way F1 hybrids. The parents and their hybrids were evaluated under two water regimes viz., normal irrigated and water stress. A total of twelve important plant descriptors were studied over a period of two years. The significant differences were observed between parents and hybrids in both water regimes. Hybrids were higher in average values for all the descriptors than parents. The role of female parent was more prominent in the expression of traits in hybrids as compared to male parents. The CMS sources varied significantly regarding seed yield per plant and other physiological traits. Proline content was three times higher in parents and their hybrids under water stress, and it was not correlated with any other descriptor. Accession CMS-PKU-2A was identified as the best general combiner for leaf area and specific leaf weight. Whereas, CMS-234A was the best general combiner for biological yield and photosynthetic efficiency under both the conditions. The cross combinations CMS-ARG-2A x RCR-8297, CMS-234A x P124R, and CMS-38A x P124R were found significant for biological yield, seed yield and oil content under both environments. Overall, this study provides useful information about the cytoplasmic effects on important sunflower traits and drought stress tolerance when used in the different combinations.

Diallel Analysis for Morphological and Biochemical Traits in Tomato Cultivated Under the Influence of Tomato Leaf Curl Virus

Eloquent information about the genetic basis of inheritance is important for any breeding program. Therefore, a diallel study was conducted under the influence of tomato leaf curl virus (TLCV) disease, using the eleven advanced lines of tomato. Firstly, the information regarding percent disease index (PDI) was determined via artificial screening with viruliferous whiteflies. Later, these lines were crossed in half diallel mating design to produce fifty-five one-way hybrids. These hybrids and parental genotypes were evaluated for seven morphological and three biochemical traits under open field conditions. Using the Griffing approach (Method II and Model I) basis of inheritance of traits were determined. Also, a Bayesian model was applied to the total yield descriptor. Correlations data indicated that total yield was not correlated with any other trait. The significant general combining ability (GCA) and specific combining ability (SCA) values indicates exploitable genetic variation. The broad-sense heritability values were larger than narrow-sense heritability, showing that selection will be efficient for the improvement of these traits. Hybrid combinations H23, H42 and H49 can be considered efficient for the selection of multiple traits, including yield. Overall, this study provides a useful information regarding the genetics of important traits of tomato under TLCV infestation.

Transcriptome Analysis of Bael (Aegle marmelos L.) a Member of Family Rutaceae

Aegle marmelos is a medicinally and horticulturally important tree member of the family Rutaceae. It is native to India where it is also known as Bael. Despite its importance; the genomic resources of this plant are scarce. This study presented the first-ever report of expressed transcripts in the leaves of Aegle marmelos. A total of 133,616 contigs were assembled to 46,335 unigenes with the minimum and maximum lengths of 201 and 14,853 bp. There were 7002 transcription factors and 94,479 simple sequence repeat (SSR) markers. The A. marmelos transcripts were also annotated based on information from other members of Rutaceae; namely Citrus clementine and Citrus sinensis. A total of 482 transcripts were annotated as cytochrome p450s (CYPs) and 314 transcripts were annotated as glucosyltransferases (GTs). In the A. marmelos leaves the monoterpenoid biosynthesis pathway was predominant. This study provides an important genomic resource along with useful information about A. marmelos.

Protein Profiling In Cancer Cell Lines And Tumor Tissue Using Reverse Phase Protein Arrays

Reverse phase protein array (RPPA) technology is an antibody-based high-throughput assay for protein profiling of biological specimens that allows for many measurements with very small amounts of cell lysate. Here, we report the sensitivity, reproducibility, and accuracy of a particular RPPA platform called Zeptosens. We customized the RPPA protocol for our in-house setup, and measured more than 80 total protein and phospho-protein levels in various cancer samples, including cell lines, organoids, tumor chunks, core needle biopsies, and laser-capture microdissected tissue samples. We discuss pros and cons of the RPPA platform, and describe results from profiling 15 cancer cell line cells using RPPA.
