Resistance screening and in-silico characterization of cloned novel RGA from multi race resistant Lentil germplasm against Fusarium wilt (Fusarium oxysporum f. sp. lentis)

Fusarium wilt caused by Fusarium oxysporum f. sp. lentis (Fol) is the most devastating disease of lentil present worldwide and in India. Identification of multi-race fusarium wilt resistance genes and incorporation into existing cultivar will help to reduce yield loss. In the present study, a hundred lentil germplasm were screened against seven prevalent races of Fol and accession IC201561, EC714243 and EC718238 were identified resistant. The typical R gene codes for the nucleotide-binding site and leucine-rich repeats (NBS-LRR) at the C terminal linked to either Toll/interleukin 1-like receptor (TIR) or coiled-coil (CC) at the N terminal. In the present study degenerate primers designed from the NBS region amplifying P-loop to GLPLA motif isolated forty-five resistance gene analogues (RGA) from identified resistant accessions. The sequence alignment identified both classes of RGA, TIR and non-TIR based on the presence of Aspartate (D) and Tryptophan (W) at the end of kinase motif respectively. The phylogenetic analysis grouped RGA into six classes, LRGA1 to LRGA6 determining the diversity of RGA present in the host. Grouping of RGA identified from Lens nigricans, LnRGA 2, 9, 13 with I2 reveals a probable role in Fusarium resistance. The similarity index of 27.85% to 86.98% was found among RGA and 26.83% to 49.41% between known R genes, I2, Gpa2, M and L6. Active binding sites present along the conserved motifs have grouped the RGA into 13 groups. ADP/ATP being the potential ligand determines ATP binding and ATP hydrolysis activity of RGA. The isolated RGA can be used in developing marker linked to the functional R gene. Further, expression analysis and full-length gene isolation further pave path to identifying the molecular mechanism involved in resistance.

Sesamol and its derivative investigated as antiandrogen - A potential prevention to prostate cancer in rats

Androgen signaling is essential for the development of prostate cancer (PCa) initiated from prostatic basal cells with collocation of androgen receptor gene mutations. Phytoestrogens, the naturally occurring compounds are AR antagonist. These compounds downregulate prostate-specific antigen (PSA) expression and cell proliferation. Thus, this gives a track to research these compounds as a possible treatment for PCa. In this work, STITCH and molecular docking predict the conformation of ligands inside the suitable target binding site. Therefore, a study was planned to know the interactions among SM and its derivatives with AR. It was further, evaluated for in vitro evaluation on LNCaP, PC-3, and DU-145 using MTT studies. The two lead compounds shortlisted from MTT studies were further analyzed for androgen-regulated genes by using RT-PCR, western blot studies and an animal model of prostate cancer. We found that SM and its derivative (3-MA) may prevent the development of PCa by androgen pathway.

Id proteins promote a cancer stem cell phenotype in triple negative breast cancer via Robo1-dependent c-Myc activation

Breast cancers display phenotypic and functional heterogeneity and several lines of evidence support the existence of cancer stem cells (CSCs) in certain breast cancers, a minor population of cells capable of tumor initiation and metastatic dissemination. Identifying factors that regulate the CSC phenotype is therefore important for developing strategies to treat metastatic disease. The Inhibitor of Differentiation Protein 1 (Id1) and its closely related family member Inhibitor of Differentiation 3 (Id3) (collectively termed Id) are expressed by a diversity of stem cells and are required for metastatic dissemination in experimental models of breast cancer. In this study, we show that ID1 is expressed in rare neoplastic cells within ER-negative breast cancers. To address the function of Id1 expressing cells within tumors, we developed two independent murine models of Triple Negative Breast Cancer (TNBC) in which a genetic reporter permitted the prospective isolation of Id1+ cells. Id1+ cells are enriched for self-renewal in tumorsphere assays in vitro and for tumor initiation in vivo. Conversely, depletion of Id1 and Id3 in the 4T1 murine model of TNBC demonstrates that Id1/3 are required for cell proliferation and self-renewal in vitro, as well as primary tumor growth and metastatic colonization of the lung in vivo. Using combined bioinformatic analysis, we have defined a novel mechanism of Id protein function via negative regulation of the Roundabout Axon Guidance Receptor Homolog 1 (Robo1) leading to activation of a Myc transcriptional programme.
