Phytochemical and antibacterial analyses of Onychium japonicum (Thunb.) Kunze.

In clinical practice bacterial resistance against antibiotics has become a serious health problem, thus using alternative approaches such as natural products as the supplementary drug could solve it. Therefore, the current study was conducted to thoroughly investigate the enrichment of phytochemicals and antibacterial potential of O. japonicum. The fronds samples of O. japonicum prepared in different solvents were used against MDR bacterial strains and phytochemical analyses. The analyses of data revealed that O. japonicum was enriched with flavonoids, alkaloids, saponins, tannins, glycosides, carotenoids, terpenoids, phlobatanins, phenols, and coumarins while quantitatively this plant has a significantly higher content of phenols(1100.91{micro}M/g) followed by sugar contents (748.67{micro}M/ml), ascorbic acid (426.12mM/g), flavonoids (160.65mg/g), anthocyanin (101.06{micro}M/g) and proline (80.58{micro}M/g). On the other hand, the organic extracts of O. japonicum were highly active against all bacterial strains while hydric extract was inactive against selected bacteria. Specifically, O. japonicum was highly active against S. aureus in all organic extracts (chloroform=16.66{+/-}0.33, ethyl acetate=15{+/-}0.57, methyl alcohol= 14{+/-}0.57, N-hexane=20.33{+/-}0.33) followed by K. pneumonia (chloroform=14.33{+/-}0.33, ethyl acetate=4.33{+/-}4.33, methyl alcohol=3.66{+/-}3.66, N-hexane=16.66{+/-}0.33) and P. aeruginosa (chloroform= 8.33{+/-}4.17, ethyl acetate=8.33{+/-}4.17, methyl alcohol=6{+/-}3.00, N-hexane=9.33{+/-}4.66), while E. coli (chloroform=0{+/-}0.00, ethyl acetate=7{+/-}3.51, methyl alcohol=3.33{+/-}3.33, N-hexane=4{+/-}4.00). Based on current findings it is concluded that O. japonicum is enriched in many useful phytochemicals that could be use as a supplement with other traditionally used antibiotics.

Structural Basis of SARS-CoV-2 Spike Protein Priming by TMPRSS2

Entry of SARS-CoV-2, etiological agent of COVID-19, in the host cell is driven by the interaction of its spike protein with human ACE2 receptor and a serine protease, TMPRSS2. Although complex between SARS-CoV-2 spike protein and ACE2 has been structurally resolved, the molecular details of the SARS-CoV-2 and TMPRSS2 complex are still elusive. TMPRSS2 is responsible for priming of the viral spike protein that entails cleavage of the spike protein at two potential sites, Arg685/Ser686 and Arg815/Ser816. The present study aims to investigate the conformational details of complex between TMPRSS2 and SARS-CoV-2 spike protein, in order to discern the finer details of the priming of viral spike and to point candidate drug targets. Briefly, full length structural model of TMPRSS2 was developed and docked against the resolved structure of SARS-CoV-2 spike protein with directional restraints of both cleavage sites. The docking simulations showed that TMPRSS2 interacts with the two different loops of SARS-CoV-2 spike protein, each containing different cleavage sites. Key functional residues of TMPRSS2 (His296, Ser441 and Ser460) were found to interact with immediate flanking residues of cleavage sites of SARS-CoV-2 spike protein. Compared to the N-terminal cleavage site (Arg685/Ser686), TMPRSS2 region that interact with C-terminal cleavage site (Arg815/Ser816) of the SARS-CoV-2 spike protein was predicted as relatively more druggable. In summary, the present study provide structural characteristics of molecular complex between human TMPRSS2 and SARS-CoV-2 spike protein and points to the candidate drug targets that could further be exploited to direct structure base drug designing.

Green Fabricated Zinc Oxide Nanoformulated Media Enhanced Callus Induction and Regeneration Dynamics of Panicum virgatum L.

The current study was focused on the usage of bio synthesized zinc oxide nanoparticles to increase the tissue culture efficiency of important forage grass Panicum virgatum. Zinc being a micronutrient enhanced the callogenesis and regeneration efficiency of Panicum virgatum at different concentrations. Here, we synthesized zinc oxide nanoparticles through Cymbopogon citratus leaves extract to evaluate the influence of zinc oxide nanoparticles on the quality of plant regeneration in switchgrass. X-ray diffraction (XRD) and attenuated total reflectance-Fourier transform infrared (ATR-FTIR) validate phase purity of green synthesize Zinc oxide nanoparticles whereas, electron microscopy (SEM) has illustrated the average size of particle 50{+/-}4 nm with hexagonal rod like shape. Energy dispersive Xray (EDS) spectra depict major peaks of Zn (92.68%) while minor peaks refer to Oxygen (7.32%). ZnO NPs demonstrate the incredibly promising results against callogenesis. Biosynthesized ZnO NPs at optimum concentration showed very promising effect on plant regeneration ability. Both the explants, seeds and nodes used in study showed dose dependent response and upon high doses exceeding 40 mg/L the results were recorded negative, whereas at 30 mg/L both explants demonstrate 70 % and 76 % regeneration frequency. The results conclude that zinc oxide nanoparticles enhance plant growth and development. Being one of the essential plant nutrients, ZnO has greatly tailored the nutritive properties at nano-scale.
