Pro-inflammatory cytokines disrupt β-cell circadian clocks in diabetes

Intrinsic {beta}-cell circadian clocks are a prerequisite for the control of glucose homeostasis through regulation of {beta}-cell function and turnover. However, little is known about the contributions of circadian clock disruption to the natural progression of {beta}-cell failure in diabetes. To address this, we examined the effects of cytokine-mediated inflammation, common to the pathophysiology of Type 1 and Type 2 diabetes, on the physiological, molecular, and epigenetic regulation of circadian clocks in {beta}-cells. Specifically, we provide evidence that the key diabetogenic cytokine IL-1{beta} disrupts functionality of the {beta}-cell circadian clock and circadian regulation of insulin secretion through impaired expression of the key transcription factor Bmal1, evident at the level of promoter activation, mRNA, and protein expression. Additionally, IL-1{beta}-mediated inflammation was shown to augment genome-wide DNA-binding patterns of Bmal1 (and its heterodimer, Clock) in {beta}-cells towards binding sites in the proximity of genes annotated to pathways regulating {beta}-cell apoptosis, inflammation, and dedifferentiation. Finally, we identified that the development of hyperglycemia in humans is associated with compromised {beta}-cell BMAL1 expression suggestive of a causative link between circadian clock disruption and {beta}-cell failure in diabetes.

Evaluation of the Luminex ARIES(R) System for the Detection and Quantification of BK virus DNA in Plasma and Serum Samples from Kidney Transplant Recipients

BKV nephropathy is as serious complication in renal transplant recipients due to the need for immunosuppression. Nearly 50% of renal transplant patients with BKV nephropathy experience a significant loss of function of the transplanted kidney. It is routine practice to screen renal transplant recipients regularly for BK viremi a. In this study we compared the performance of BKV qPCR analyte specific reagents (ASR) by ELITech, and Luminex for measuring BKV viral load in plasma and serum using the Roche Cobas(R) z480 instrument, and the Luminex ARIES(R) platform respectively. A total of 34 patients previously tested on the z480, with results spanning the test s linear range, were analyzed on the ARIES(R). The BKV DNA copy number correlation between the two methods was very good with an R2 value of 0.98. The average difference in log copy number between the two methods was -0.3, indicating that the ARIES(R) method may have slightly greater analytical sensitivity. BKV quantification results were closely matched between the two different methods. The workflow with the ARIES(R) System is greatly simplified by elimination of DNA extraction and most hands-on steps. The high degree of automation allows samples to be tested as they arrive into the laboratory, resulting in enhanced patient care due to more rapid turnaround time for results back to the ordering physician.
