Excitation-contraction coupling plasticity is revealed upon RyR2 depletion in adult mouse ventricular myocytes

Ryanodine receptor 2 (RyR2) is central to cardiac excitation-contraction coupling (ECC), translating electrical signals into sarcoplasmic reticulum (SR) Ca2+ release and mechanical contraction. Although constitutive deletion of RyR2 is lethal, adult hearts can withstand a significant reduction in RyR2 expression without overt ECC dysfunction. We hypothesized that ECC possesses intrinsic homeostatic plasticity that rapidly adapts to acute RyR2 loss to maintain cardiac performance. Using an adult tamoxifen-inducible RyR2 conditional knockout (RyR2cKO) mouse model, we evaluated cardiac structure, electrophysiology, and intracellular Ca2+ dynamics. RyR2cKO mice survived for weeks with <10% residual RyR2 protein expression. In vivo echocardiography and electrocardiography revealed normal cardiac structure and function. Confocal Ca2+ imaging in intact hearts and isolated ventricular myocytes showed preserved Ca2+ transient (CaT) amplitude but slower release kinetics and elevated SR load. Simultaneous patch-clamp and confocal Ca2+ imaging established that L-type Ca2+ current (ICa) and ECC gain were maintained in RyR2cKO myocytes. Expression of CaV1.2, NCX, and phospholamban remained unchanged, whereas SERCA2a, IP3 receptor, and VMP1 levels decreased. Pharmacological inhibition of IP3 receptors did not suppress CaT, ruling out alternative SR channel compensation. These findings reveal remarkable intrinsic ECC plasticity, where high RyR2 channel reserve, elevated SR Ca2+ load, and CaT autoregulation collectively maintain cardiac Ca2+ cycling and contractile function despite profound RyR2 depletion.

A large, comprehensive labeled MRI dataset of the human lateral ventricle choroid plexus

The choroid plexus (ChP) is a highly vascularized structure located in the brain ventricles that plays important roles in multiple central nervous system functions. ChP enlargement has been implicated in many neuropsychiatric conditions such as multiple sclerosis, depression, schizophrenia, and Alzheimer's disease. However, limitation in the accuracy of the current segmentation tools highlight the need for more reliable methods to quantify ChP volume in healthy and clinical human populations. The shape and size of the ChP vary drastically from person to person, and currently there are no large ChP datasets which allow comprehensive training of segmentation models. Here we introduce DataPlex, a collection of 700 structural MRI brain images where the lateral ventricle ChP has been manually labeled and checked by expert human raters. DataPlex is balanced and representative across a wide range of scanning parameters, demographics, and clinical profiles, and subject-level information will be available for each labeled image. DataPlex will be open source to support the creation of reliable segmentation tools for the human lateral ventricle ChP.

Over-expression of the photoperiod response regulator ZmCCT10 modifies plant architecture, flowering time and inflorescence morphology in maize

Maize originated as a tropical plant that required short days to transition from vegetative to reproductive development. ZmCCT10 [CO, CONSTANS, CO-LIKE and TIMING OF CAB1 (CCT) transcription factor family] is a photoperiod regulator and was identified as a major QTL controlling photoperiod sensitivity in maize. We modulated expression of ZmCCT10 in transgenic maize using two constitutive promoters which cause differing expression levels. Transgenic plants over expressing ZmCCT10 with either promoter was delayed in their transition from vegetative to reproductive development but were not affected in their change from juvenile-to-adult vegetative growth. Strikingly, transgenic plants containing the stronger expressing construct had a very prolonged period of vegetative growth accompanied with dramatic modifications to plant architecture that impacted both vegetative and reproductive traits. These plants did not produced ears, but tassels were heavily branched, and more than half of the transgenic plants showed conversion of shoot apices into \"bushy tops\", which were composed of vegetative reversion plantlets. Analysis of expression modules controlling the floral transition and meristem identity linked these networks to photoperiod dependent regulation, whereas phase change modules appeared to be photoperiod independent. Results from this study clarified the influence of the photoperiod pathway on vegetative and reproductive development and allowed to fine-tune the flowering time model for maize.
