Identification of a Novel Spliced Variant of ADAMTS13 in Human Mesenchymal Stem Cells

ADAMTS13 is a matrix metalloproteinase that cleaves von Willebrand factor (vWF) into small multimers. Several truncated forms of ADAMTS13 can be detected in plasma, generated due to alternative splicing or proteolysis by serine proteases. In this study, for the first time, we report an alternatively spliced variant of ADAMTS13 that is expressed in human mesenchymal stem cells (MSCs), derived from both the Whartons Jelly (WJ) of umbilical cords and the decidua-basalis layer of the placenta (PL). Our results demonstrated that the variant contained the signal peptide (Sp), propeptide (P), metalloprotease (Mp), disintegrin-like (Dis), thrombospondin type-1 repeat 1 (Tsp1-1) and cysteine-rich (Cys) domains, while the spacer (Spc) and CUB domains were absent. The Tsp1-2-8 domain was partially present. Additionally, this variant possessed partial vWF cleavage activity compared to full-length ADAMTS13. Thus, we identified a novel isoform of ADAMTS13, generated due to exon skipping in human MSCs, which retained partial activity towards vWF cleavage.

Smartphone-assisted real-time estimation of chlorophyll and carotenoid contents in spinach following the inversion of red and green color features

PurposeChlorophyll (Chl) content is a reliable indicator of leaf nitrogen content and plant health status. Currently available methods for image-based Chl estimation require complex mathematical derivations and high-throughput imaging set-up along with multiplex image-preprocessing steps. Further, the influence of carotenoid (CAR) content has been largely ignored in the process. The present study describes a smartphone-based leaf image analysis method for real-time estimation of Chl content and Chl/CAR ratio. MethodsColor features were obtained from RGB (red, green, blue) images of spinach leaves using a smartphone, and inverse R and G values were calculated. Correlation analysis of color indices and photosynthetic pigment (PP) contents was performed, followed by principal component analysis (PCA). Linear mathematical modeling was performed for describing regression equations for predicting PP contents. Results1/R and 1/G showed strong positive linear correlation (0.93 < r2 < 0.96) with Chl and CAR contents, respectively. Furthermore, 1/R+1/G and [1/R]/[1/G] presented strong positive linear correlation with Chl + CAR (r2 = 0.95) and Chl/CAR (r2 = 0.88), respectively. PCA confirmed the association of color indices with the respective PP features, which were subsequently estimated using the correlation models. A smartphone-based companion application was developed using the linear models for non-invasive, real-time estimation of Chl content and Chl/CAR ratio. ConclusionThe ratios 1/R and 1/G indicate the contents of Chl and CAR via linear models. The smartphone application developed using the linear models enables real-time estimation of Chl content and Chl/CAR ratio without complicated image preprocessing steps or mathematical derivations.

Down-regulated GDF9 and BMP15 mRNAs are the important determining factors for the unexplained female infertility cases advised for assisted reproductive technology

BackgroundIn India, the potential risk factors influencing female infertility are not well-characterized. Hence, this study was initially aimed to determine the multi-factor risk-predictive models for the infertile females from different Indian ethnics. Afterwards, when treatment was performed for correcting the abnormal demographic and clinical parameters obtained from our initial findings, some of the patients were observed to be treatment-non-responsive. This further recommended assessing the expression pattern of GDF9 and BMP15 mRNAs in those unexplained infertile females. MethodologyThis study included 470 Primary, 209 Secondary infertile and 419 age-matched Healthy females, where baseline demographic and clinicopathological factors were assessed. Additionally, from the treatment-non-responsive patients, laparoscopy and trans-vaginal ultrasound-guided collection of ovarian follicular fluid were done for studying the expression pattern of GDF9 and BMP15 mRNAs using q-PCR. Principal FindingsMid-education level, no contraception usage and duration of infertility for >2 years were the critical risk factors for female infertility (OR>>1; p<0.05), whereas elevated platelet count showed ~10-fold more risk of having secondary infertility and high ESR showed 3.33-fold increased threat of primary infertility. Besides, abnormal MCV, LH/FSH ratio, mild-anaemia and hypothyroidism were the best risk-predictive multi-factor model. After correcting these abnormal clinical factors, 6.8% of the patients were treatment-non-responsive, where elevated ESR, hypothyroidism and anaemia remained uncorrected. Interestingly, in those unexplained cases, GDF9 and BMP15 mRNAs were significantly down-regulated (2-4 fold) irrespective of age groups. Additionally, for predicting female infertility, the cut-off for the expression level of GDF9 and BMP15 mRNAs were estimated -2.09 and -2.75, respectively, with a relatively high sensitivity ([≥]62.5%) and specificity (75%). ConclusionsGDF9 and BMP15 mRNAs may be considered as molecular markers for predicting female infertility. Their down-regulation along with hypothyroidism and anaemia probably led to the treatment-non-responsiveness, thus, those patients may be advised to assisted reproductive technology with supplementation of exogenous GDF9 and/or BMP15.
