Development and validation of KASP markers for a novel powdery mildew resistance gene in wheat using BSR-Seq analysis

Wheat powdery mildew occurs as a devastating infection caused by the pathogenic fungi Blumeeria graminis f. sp. tritici (Bgt). The management of disease becomes much more effective when host resistance methods are employed. The WL711-2U/2B line served as the introgressed line to identify genes linked with powdery mildew resistance. This allowed us to look into the potential resistance components in response to powdery mildew infection. The resistance status of the line towards powdery mildew developed from a single dominant gene named PmAT. Bulked segregant RNA-Seq (BSR-Seq) together with previous QTL-Seq SNP markers helped identify strong candidate regions in chromosome arm 2B extending from 531114848 bp to 530235618 bp. The LOD score from this region reached 72.4 with a PVE% of 64.7, thus proving its main role in resistance. The physical position was subsequently locked up, which was different from the previously identified genes on the same chromosome arm in its position, suggesting that it is most likely a new Pm gene. The QTL interval contained five potential genes that we identified during this research. The five candidate genes directs the protein production, which performs functions linked to the pentatricopeptide repeat family as well as B-box-type zinc finger domain, P-loop-containing nucleoside triphosphate hydrolase, and plant peroxidase. HighlightThe identification of a new wheat powdery mildew resistance gene (PmAT) using a combination of BSR-Seq and QTL-Seq analysis, which located five candidate genes specifically on chromosome 2B.

Role of the transcriptional regulator SP140 in resistance to bacterial infections via repression of type I interferons

Type I interferons (IFNs) are essential for anti-viral immunity, but often impair protective immune responses during bacterial infections. An important question is how type I IFNs are strongly induced during viral infections, and yet are appropriately restrained during bacterial infections. The Super susceptibility to tuberculosis 1 (Sst1) locus in mice confers resistance to diverse bacterial infections. Here we provide evidence that Sp140 is a gene encoded within the Sst1 locus that represses type I IFN transcription during bacterial infections. We generated Sp140-/- mice and find they are susceptible to infection by Legionella pneumophila and Mycobacterium tuberculosis. Susceptibility of Sp140-/- mice to bacterial infection was rescued by crosses to mice lacking the type I IFN receptor (Ifnar-/-). Our results implicate Sp140 as an important negative regulator of type I IFNs that is essential for resistance to bacterial infections. Impact StatementRepression of type I interferons by SP140 is essential for resistance to Legionella pneumophila and Mycobacterium tuberculosis.

Identification, expression analysis and molecular modeling of Iron deficiency specific clone 3 (Ids3) like gene in hexaploid wheat

Graminaceous plants secrete iron (Fe) chelators called mugineic acid family phytosiderophores (MAs) from their roots for solubilisation and mobilization of unavailable ferric (Fe3+) ions from the soil. The hydroxylated forms of these phytosiderophores have been found more efficient in chelation and subsequent uptake of minerals from soil which are available in very small quantities. The genes responsible for hydroxylation of phytosiderophores have been recognized as iron deficiency-specific clone 2 (Ids2) and iron deficiency-specific clone 3 (Ids3) in barley but their presence is not reported earlier in hexaploid wheat. Hence, the present investigation was done with the aim:(i) to search for the putative Hordeum vulgare Ids3 (HvIds3) ortholog in hexaploid wheat, (ii) physical mapping of HvIds3 ortholog on wheat chromosome using cytogenetic stocks developed in the background of wheat cultivar Chinese Spring and (iii) to analyze the effect of iron starvation on the expression pattern of this ortholog at transcription level. In the present investigation, a putative ortholog of HvIds3 gene was identified in hexaploid wheat using different bioinformatics tools. Further, protein structure of TaIDS3 was modelled using homology modeling and also evaluated modelled structure behavior on nanoseconds using molecular dynamics based approach. Additionally, the ProFunc results also predict the functional similarity between the proteins of HvIds3 and its wheat ortholog (TaIds3). The physical mapping study with the use of cytogenetic stocks confines TaIds3 in the telomeric region of chromosome 7AS which supports the results obtained by bioinformatics analysis. The relative expression analysis of TaIds3 indicated that the detectable expression of TaIds3 induces after 5th day of Fe-starvation and increases gradually up to 15th day and thereafter decreases till 35th day of Fe-starvation. This reflects that Fe deficiency directly regulates the induction of TaIds3 in the roots of hexaploid wheat.
